Для работы с СОМ в общем-то достаточно функций
::CreateFile(), ::ReadFile(), ::WriteFile(), ::CloseHandle(), ::SetCommState(), ::SetCommTimeouts()
ну и структур DCB и COMMTIMEOUTS. Описания есть в MSDN
